Error corrected Duplex Sequencing of Sprague Dawley rat liver treated with benzo[a]pyrene or N-ethyl-N-nitrosourea

Duplex Sequencing was applied to DNA samples extracted from the livers of untreated Sprague Dawley rats or rats treated with either 100mg/kg/day benzo[a]pyrene (BaP) for ten day or 40 mg/kg/day N-ethyl-N-nitrosourea (ENU) for three days. Using the measured mutation frequency (MF) in these original samples, mixtures were constructed to create standards with target 1.2-, 1.5-, and 2-fold greater MF than the untreated control. Aliquots of these standards were distributed to eight laboratories in North America and Europe. Duplex Sequencing libraries were prepared by each laboratory.
