To identified TS p53 mutations and PAT-treatable p53 mutations in mammalian U937 cells. Overall design: 827 plasmids encoding p53 with either 815 frequent missense mutations, 6 truncation mutations (E11stop, P98stop, P151stop, P152stop, A159stop, P177stop), or 6 synonymous mutations (P151P, G154G, A159A, P177P, G244G, R282R) were stably expressed in U937 cells. This cell library was treated with 0, 5, 10 µM PAT or sub-physiological temperature (32 ?) for 7 days, followed by p53 variant abundance using next-generation sequencing.
