We have employed a single-cell sequencing approach using 10x Genomics scRNAseq to study the impact of Disc1 locus impairment (LI) on adult microglia. This study revealed that actin dysregulation and enhanced phagocytosis characterize the transcriptomic profile of Disc1 LI microglia. CD45lowCD11b+ immune cells were sorted out of the brains of 23-week-old homozygous mutated (Disc1 LI/LI) and wild-type (Disc1 WT/WT) littermate controls (n = 6 males per group) using a BD FACS ARIA II. Cells were analyzed using scRNAseq.
