Group B Streptococcus transposon-sequencing screen in calprotectin

To determine the genes important for survival in calprotectin, we performed high-throughput transposon sequencing. Briefly, a saturated transposon mutant library was exposed to subinhibitory and inhibitory concentrations of calprotectin, following incubation cultures were plated on media selecting for transposon insertions. DNA was extracted from the input and output pools, libraries were prepared, and Illumina sequencing was performed.
