Whole transcript analysis of amyloid beta 42 (Aβ42)-induced SH-SY5Y cells in control and treated groups (curcumin, piperine and combination therapy) were assessed using microarray profiling. A number of up-regulated and down-regulated genes were altered in sample-specific group. In this study, an optimized concentration of 35 µM of curcumin and piperine in combination was used to treat Aβ42 fibril and then high-throughput microarray profiling (Clariom S assay) was performed on the extracted RNA from pelleted cells. Twelve total RNA samples were submitted to UMBI ("the service lab"). All the RNA samples passed initial QC checks and were determined to be of good quality to continue with whole transcripts analysis. All samples were processed according to Applied Biosystems™ recommended protocols, and the service lab Standard Operating Procedures (SOP). With the consent from the customer, all 12 samples which passed the assay quality control (QC) check points were selected and hybridized to Human Clariom S Array. The data generated passed all our quality metrics with POS vs NEG AUC of values of above 0.9.
