Comparative analysis of physilogy and wax components showed that a low-wax mutant exhibited weaker drought resistance with different wax compositions. Comparative analysis of transcriptome and cuticular lipidome, some critical candidate genes involved in alkane synthesis and alcohol synthesis of wax synthesis pathway were identified such as CER1, CER1-L, CER3, FAR3, MAH1. Besides, a possible pathway of ketone synthesis was discovered that BnaA02g17240D (MAH1) directly catalyzed VLC alkanes into VLC ketone. We also found that a nucleotide mutation in the second exon and the NAD(P)-binding rossmann-like domain of BnaA01g03200D (FAR3), a critical candidate genes of alcohol synthesis in the low-wax mutant, resulted in the converation of glutamate to aspartate.
