Due to their unique longevity and capacity to secrete high levels of protein, plasma B cells play have the potential to be used as a cell therapy for protein replacement. Here, we show that ex vivo engineered human plasma cells exhibited transcriptional features of long-lived plasma cells. Overall design: Ex vivo culture human B cell with proliferation and differentiation stimulants. Followed by CD138 isolation and CD38 isolation at day13 (D11), collecting isolated cell and keep culture this two groups (CD138+ vs CD38+) of cells. After 6 days, harvest these two groups of cells at day19(D17). All the collected cells are put in trizol and store in -20.
