Streptococcus pneumoniae is the leading cause of community-acquired pneumonia. The release of the pore-forming cholesterol-dependent cytolysin (CDC) pneumolysin (PLY) and hydrogen peroxide (H2O2), a physiological metabolite, is an important virulence determinant of pneumococci. Overall design: To investigate the impact of endogenous pneumococcal hydrogen peroxide on the activity and release of pneumolysin we grew the bacterial strains, generated PLYC428S and isolated PLY, determined hemolitic activity, detected and quantified PLY, performed adhesion assay, as well as RNA sequencing.
