RNAseq of MLL-AF9+ lin- ckitlo sca1- (LKlo) cells was performed after sorting these cells from the BM of Nes-creERT2;iDTA (N=8) or control (N=6) mice.

Gene expression changes in leukemic LKlow cells after depleting nestin+ cells in vivo Sorted cells were combined for a total of 3 biological replicates (each consisting of pooled LKlo cells from from 2-3 mice). Total RNA was isolated using the Arcturus Picopure RNA Isolation Kit (Thermo Fisher Scientific). RNA was amplified and prepared for RNA-Seq using the Ovation RNA-Seq System v2 (NuGEN) following the manufacturer’s recommendations.
