By genome-wide ChIP-seq analysis in ABC DLBCL cell lines, we identified 2,251 STAT3 direct target genes, which involve B cell activation, survival, proliferation, differentiation and migration. Whole transcriptome profiling revealed that STAT3 acts as both a transcriptional activator and suppressor. TMD8 and OCI-LY10 cells were treated by eithor DMSO or AZD1480 (JAK inhibitor) for 4hrs, ChIP-seq experiments were performed by STAT3 antibody (sc-482X, Santa Cruz Biotechnology). Cells transduced with control shRNA, shSTAT3#10 or shSTAT3#16 in TMD8 and OCI-LY10 cells were induced with 20 ng/ml doxycycline for 2 days. Total RNA from these cells was then used for RNA-seq analysis.
