The selective 2'-hydroxyl acylation analyzed by primer extension and mutational profiling (SHAPE-MaP) approach was applied to investigate the RNA secondary structure surrounding uORF2 and its flanking sequences in the TPM3P9 transcript. Cells were treated with DMSO (control) or 1M7 to probe nucleotide flexibility. The resulting libraries were barcoded, pooled, and sequenced on the Illumina NovaSeq 6000 platform using paired-end 250 bp reads. The dataset aims to elucidate how local RNA structural features around uORF2 may influence osteolysin translation in bone-metastatic breast cancer cells.
