DNA methylation plays a major role in the epigenetic regulation of gene expression and is essential for developmental potential of the cell. Although a few DNA methylation pro?ling studies of porcine genome which is one of the important biomedical models for human diseases have been reported, the available data are still limited.The aim of the project was to conduct genome-scale DNA methylation profiling at nucleotide resolution of adipose tissue mesenchymal stem cells (AMSCs), bone marrow MSC (bmMSC), adipose derived MSC (adMSC), porcine fetal fibroblast (pFF) and kidney derived fibroblasts (KDNF), that displayed high and low efficiency for somatic cell nuclear transfer (SCNT).Cell lines were isolated from adipose tissue, kidney and fetal fibroblast of adult male pigs (n=17; high efficiency cells=7; low efficiency cells= 10 ) according to isolation protocol. Samples named 2,5,9,12, 28, 29, 30 display high efficiency, and samples named 14,15,16,17,18,19,21,22,23,26 display low efficiency cells for SCNT. The cell line was maintained according to standard protocol. Genomic DNA was extracted from the cell lines using standard phenol:chloroform extraction.
