Experience-dependent gene expression reshapes neural circuits, permitting learning of knowledge and skills. However, the diversity of plasticity-related transcriptional responses across neurons underlying learning remains poorly understood. Here, we analyzed single-nucleus transcriptomes of L2/3 glutamatergic neurons of the primary motor cortex after motor skill training or home-cage control in water-restricted mice. Overall design: The caudal forelimb area of primary motor cortex for 6 mice was analyzed using snRNAseq (shallow and deep sequencing was performed for each mouse). 3 mice were control, and 3 were trained on a lever-press task. barcode suffixes (-1 to -6) correspond to the samples titled "M1 nuclei, control, mouse1, shallow snRNAseq ", "...mouse2...", "â¦mouse3â¦", etc., respectively
