In this paper, we show that Csnk1a1 was highly expressed in Sertoli cells and the conditional knockout of Csnk1a1 in murine Sertoli cells leads to severe testicular atrophy and infertility, characterized by rapid Sertoli cells depletion and spermatogenesis dysfunction, and infer that Csnk1a1 is required for the fate control of fetal and neonatal Sertoli cells of mice. At molecular levels, we found Csnk1a1 interacted with a splicing factor ZRSR1 to regulate the apoptosis of Sertoli cells. These results provide new insights into the roles of Csnk1a1 in alternative splicing and male reproduction and crucial for our understanding regulating the mechanisms of testicular development and male reproduction.
