WUS is the key regulator in stem cell induction and maintenance. We performed the small RNA high throughput sequencing to test whether WUS is involved in stabilizing RNA structure in Arabidopsis thaliana. For analyzing the degradation of RNA induced by WUS, the pUBI10:mCherry-GR-linker-WUS plants sprayed with or without DEX (15μM) for 2 days, and then total RNA were isolated from the shoot apexes. 18-60 nt small RNA were selected for library building.Two biological replicates were used for small RNA library preparation. To annotate reads of small RNA from Arabidopsis, we first map clean reads to the Arabidopsis genome.
