Control of Ae. aegypti is challenging, and is further compromised through a recent rapid rise across Latin America of knockdown resistance (kdr) to pyrethroid insecticides. Widespread, heavy use of pyrethroid space sprays – due to their strong human safety profile – has created immense selection pressure for resistance. This resistance is primarily under the control of the voltage gated sodium channel gene (para), however metabolic mechanisms also contribute to resistance. In this study, 3rd generation colony mosquitoes were exposed to pyrethroid (deltamethrin, 3 ug or permethrin, 25ug) in bottle assays for one hour. Those not knocked down were designated as resistant(alive) and those that did not recover after 4 hours post-treatment were designated as susceptible("dead"). Dual replicate pools of 25 were sequenced and enriched for genic sequences. Illumina paired end reads were trimmed using cudadapt. Collection VCP- Viva Caucel, permethrin; VCD, Viva Caucel, deltamethrin, VeP, Vergel, permethrin. -VCPA permethrin-treated, resistant, replicates 1 and 2 are SRR5805471 and SRR5805472, respectively. VCPD permethrin-treated, susceptible, replicates 1 and 2 are SRR5805473 and SRR5805470, respectively. VCDA deltamethrin-treated,resistant, replicates 1 and 2 are SRR5805467 and SRR5805466, respectively. VCDD deltamethrin-treated,susceptible, replicates 1 and 2 are SRR5805469 and SRR5805468, respectively. VePA permethrin-treated, resistant, replicates 1 and 2 are SRR5805465 and SRR5805464, respectively. VePD permethrin-treated, susceptible, replicates 1 and 2 are SRR5805475 and SRR5805474, respectively.
