Characteristics of the chondroprogenitors cells remain unidentified. Here we identify a chondroprogenitor population, which specifically expresses Procr,and used single cell RNA sequencing (scRNA-seq) and bulk RNA-seq to analyze the diversity of Prcor+ cells in articular cartilage and periosteum. Overall design: Articaular cartilage cells from sham and DMM surgery mice were isolated by Fluorescence-activatedcell sorting (FACS) with APC-conjugated CD31, CD45, and Ter119 antibody and analyzed using scRNAseq. Long bone periosteum cells from Prrx1-Cre;tdTomato;Col2.3-GFP were isolated by Fluorescence-activatedcell sorting (FACS) with APC-conjugated CD31, CD45, and Ter119 antibody and analyzed using scRNAseq. Articaular cartilage cells from steady state mice were isolated for Procr+CD105- and Procr+CD105+ cells, and analyzed using RNAseq.
