We deviced a new gene editing method, PRIME-In, for targeted integration of large DNA cargos independent of introducing genomic double stranded breaks (DSBs). We evaluated the editing efficiency and fidelity of our method by PCR-enriched next generation sequencing in comparison with a traditional DSB-triggered high-efficiency integration approach (HMEJ).
