Squamocolumnar junction (SCJ) of the uterine cervix is a target of human papilloma virus (HPV) infection and thereby a putative cell-of-origin for uterine cervical cancer. However at present, in vitro models that accurately phenocopy its homeostasis is lacking. In this study, we aimed at establishing organoids of SCJ cells and identifying its characteristics. Samples were collected from the uterine cervix of 4 cases who underwent hysterectomy at our department, and processed to initiate organoid culture. Organoids were obtained from 3 samples among them, and they were analyzed both histologically and by transcriptome analysis. In this study, expression profiles of organoid cultures from uterine cervix were compared with those of three cell lines Cervical tissues were obtained from patients who underwent hysterectomy without any malignant findings on their uterus. In 3 cases, these tissues were propagated and maintained by organoid culture. Total RNA samples were extracted from these passaged samples. In comparison, RNA samples were isolated from 3 cell lines, Ect1/E6E7, End1/E6E7 and Normal immortal human keratinocytes (NIKS), which were obtained from ATCC and cultured as previously described. All of these RNA samples were subjected to TORAY 3D-gene analysis using the 3D-Gene Human Oligo chip 25K.
