This study is based on the importance of stem cells in future medicine and the need to understand the fundamental mechanisms that regulate their potential to differentiate towards specific lineages. We aim to gather the information required to understand the regulatory networks of key stem cells and more specifically, to understand the transitions between stem cell states using embryonic stem (ES) cells and a recently described intermediate, Epiblast stem (EpiS) cells. Based on RNAi drop-out screens in ES and EpiSC cells, we have selected 125 candidate genes for functional and transcriptomic analysis in ES and EpiS cells 72 hours following RNAi knockdown.
