Our work demonstrates that nuclear cholesterol plays a direct role in transcriptional regulation through its interaction with BASP1. Overall design: Investigating the role of BASP1 and its interaction with cholesterol in transcriptional regulation and chromatin reorganisation using high-throughput sequencing assays for differential gene expression and chromatin accessibility. Experiments were performed in triplicate from culture replicates for K562 cells, K562 cells expressing wild type BASP1 and K562 cells expressing the BASP1 mutant BASP1-G2A.
