Overcoming MCT8-dependent oligodendrocyte dystrophy in progressive multiple sclerosis to enhance neuroprotection and neurorepair

Secondary progressive multiple sclerosis (MS) is a stage of MS with few effective treatments, leaving a major unmet medical need. This study explored how loss of MCT8, a protein that transports thyroid hormone, contributes to MS by harming oligodendrocytes—cells that make myelin and protect nerves. Using human tissue and mouse MS models, we found that MCT8 is essential for oligodendrocyte survival and myelin repair. We tested DITPA, a synthetic thyroid hormone that bypasses MCT8, and showed it protects oligodendrocytes, reduces inflammation, and promotes remyelination, making it a promising therapeutic candidate for progressive MS.
