extracellular PTEN treatment did not affect the growth of PTEN knockout B16-F10 cells cultured in vitro. , To investigate whether extracellular PTEN act on the tumor microenvironment to exert a tumor-suppressive role in vivo,molecular changes caused by PTEN treatment inside the B16-F10-?PTEN tumors were monitored by RNA sequencing. Overall design: 1.0×105 PTEN knockout B16-F10 cells were subcutaneously injected into C57BL/6 mice.once tumors reached ~ 50-100 mm3 in volume, mice were randomly assigned to control or treatment groups and were intratumorally or intraperitoneally injected with PTEN proteins or PBS every day. tumors were harvested for scRNAseq
