The study is designed to understand the role of TET mediated demethylation and its intricate interplay with histone enhancers and loci-specific 5-hmC levels of candidate genes in breast cancer. Overall design: TET1 and TET2 depletion have been performed using siRNA-mediated gene silencing in MCF-7 breast cancer cell lines. TET-deficient and the control cells were pulled with H3k27ac and H3K4me1 antibodies followed by ChIP bead purification, library preparation, and sequencing.
