Sesame is an important oilseed crop with many pharmacological properties. We have made inter-specific hybridization between common cultivated high yielding Indian sesame (Sesamum indicum) and one of the wild sesames (S. mulayanum). The wild is low oil yielding yet unlike the cultivated sesame its oil contains very high lignan (Sesamin, Sesamolin) of anti-oxidant properties. Most of the traits of seed like oil, testa colour etc are QTL-controlled and we can expect the involvement of many genes. Our breeding population has a remarkable variation in those traits. We have performed transcriptomics of the seeds in different developmental stages from the parents and six selected recombinant lines (RILs, F6). From the RNA-Seq data, we have investigated variation in gene sequences, expression level and correlated them with trait variation. This will foster the development of candidate gene based SNP markers and help in sesame breeding programme for a better genotype.
芝麻是一类兼具多种药理活性的重要油料作物。本研究以高产的印度栽培芝麻(Sesamum indicum)与野生芝麻物种之一(S. mulayanum)开展种间杂交。该野生种含油量较低,但其油脂中富含具有抗氧化活性的木脂素(芝麻素(Sesamin)、芝麻林素(Sesamolin)),这一点与栽培芝麻截然不同。种子多数性状(如含油量、种皮颜色等)均受数量性状基因座(Quantitative Trait Locus, QTL)调控,且推测涉及多个基因的参与。我们的育种群体在上述性状上存在显著变异。本研究对亲本及6个筛选得到的重组自交系(Recombinant Inbred Lines, RILs,F6代)种子的不同发育阶段开展了转录组测序分析。基于RNA测序(RNA-Seq)数据,我们解析了基因序列与表达水平的变异,并将其与性状变异进行关联分析。本研究将助力基于候选基因的单核苷酸多态性(Single Nucleotide Polymorphism, SNP)标记开发,并可为选育优良基因型的芝麻育种工作提供支持。
