Investigation of the PAR gene conservation in Taraxacum germplasms

We investigated the conservation of the identified Taraxacum officinale PAR gene in Taraxacum germplasm by high-throughput re-sequencing using the Illumina HiSeq2500 sequencer. Five apomictic (T.richardsianum, T.albidum, T.brevicorniculatum, T.brachyglossum, T.gratum) and two sexual (T.cylleneum, T.koksaghyz) species, belonging to different sections, were grown. DNA was extracted from leaves using QIAGEN DNeasy Plant DNA extraction kit and quantified with Qubit. DNA was sonicated and PCR free Illumina sequencing libraries were prepared with an average insert size of 580-600 bp. The variable genome size of the Taraxacum germplasm was taken in account by normalizing the HiSeq library sample concentrations to obtain an average of 17 x coverage per allele utilizing eight lanes of 125bp paired end Illumina sequencing.
