Kinetic Stem Cell Counting Inter-Lab Study Data for Human Oral Alveolar Bone-Derived Mesenchymal Stem Cells

The submitted data are results from an inter-lab study conducted to evaluate the precision of the kinetic stem cell (KSC) counting method for quantifying the specific fraction of tissue stem cells in heterogeneous tissue cell preparations used for stem cell research, stem cell medicine, and pharmaceutical drug development. Replicate samples of oral alveolar bone mesenchymal tissue cells, which contain mesenchymal stem cells (MSCs), from a single human donor were used for the study. At three geographically distinct laboratory sites, three each of nine replicate samples were evaluated using the same standard operating procedure (SOP) for KSC counting. Cultures were passaged every 72 ± 3 hours with a 1:20 split basis until no cells were detectable. The reported data were obtained from triplicate serial cell cultures performed for each of the nine replicate tissue cell samples. The data include cumulative population doubling (CPD) data and corresponding dead cell fraction (DCF) data determined by trypan blue dye-exclusion. CPD data and DCF data are the inputs for the KSC counting TORTOISE Test® computational simulation software that is used to determine the stem cell-specific fraction (SCF) or dosage (SCD) of tissue cell samples. A single technician performed the SOP at each site. Each site used a different instrument for cell counting. The replicates for Site 1 and Site 2 were shipped to them on dry ice; they were of the same culture lineage; and their culture medium was supplemented with the same lot of fetal bovine serum (FBS). The replicates at Site 3 were not shipped; they were from a parallel culture lineage derived with a different lot of supplemented FBS; and the different lot of FBS was also used for their serial cultures as well. These data were developed to evaluate the intra-site and inter-site precision of input CPD data (https://www.astm.org/workitem-wk84181), of KSC counting SCF determinations (submitted), and of KSC counting derivations for rapid stem cell counting algorithms (submitted).
