We used a new technology, named Paired-Damage, to perform simultaneous profiling of DNA oxidative damage and transcriptome on Hela cells and mouse cortex at single-cell level. We also performed matched damagenome, ATAC-seq and RNA-seq of Hela cells at bulk level. We develop Paired-Damage for simultaneous profiling the genome-wide DNA oxidative damage and transcriptome, in highly scalable single-cell manner. Paired-Damage measures all the major oxidative DNA damage including 8-oxoG, AP sites and single-strand breaks, together with transcriptome from one same cell. To demonstrate the utility of Paired-Damage, we applied it in Hela cells treated with oxidative stress condition and mouse brain cortex cells. We also performed matched damagenome, ATAC-seq and RNA-seq of Hela cells at bulk level.
