Phase II study of single-agent daratumumab in patients with high-risk Monoclonal Gammopathy of Undetermined Significance (MGUS) and low-risk Smoldering Multiple Myeloma (SMM)

We conducted a Phase II study of single-agent Daratumumab in patients with high-risk Monoclonal Gammopathy of Undetermined Significance (MGUS) and low-risk Smoldering Multiple Myeloma (SMM) to determine if earlier treatment can lead to deep responses and prevent progression to MM (D-PRISM, NCT03236428, n=41). To identify genomic and immune biomarkers of response, we conducted single-cell RNA sequencing of tumor and immune cells from 36 patients enrolled in the trial.
