<p>Surface sediment samples were collected for potential nitrification rates, using sediment slurries with filtered site water. Ammonia oxidizer gene abundances (AOA and AOB <em>amoA</em>) were quantified using qPCR, and clone libraries for each gene were sequenced using Sanger sequencing.</p>

本研究采集表层沉积物样品,采用过滤后的原位水配制沉积物悬液以测定潜在硝化速率。通过实时荧光定量PCR(quantitative PCR, qPCR)定量分析氨氧化菌功能基因——氨氧化古菌(Ammonia-Oxidizing Archaea, AOA)与氨氧化细菌(Ammonia-Oxidizing Bacteria, AOB)的amoA基因——的丰度,并利用桑格测序(Sanger sequencing)完成各基因克隆文库的测序工作。
