This project consisted of isolating and sequencing antibiotic-resistant Enterococcus spp., Klebsiella spp., and Escherichia coli from multiple stages of wastewater treatment. Isolates were cultured from influent, after each stage of treatment, after each stage of disinfection, and into the distribution or groundwater recharge. Isolates were confirmed to be resistant to the selective antibiotic (cefotaxime for E. coli and Klebsiella spp., vancomycin for Enterococcus spp.) by Kirby-Bauer and tested for phenotypic multi-drug resistance. A subset of multi-drug resistant isolates varying in isolation stage were selected for whole genome sequencing.
