Boletus edulis complex in the British Isles. With B. edulis showing strong evidence of selective pressures already acting to recapitulate past speciation events, our aim is to understand how this specialisation leads to speciation at the genomic level, in terms of which genes and genomic regions most associated with adaptation. We have already assembled a comprehensive database comprising over 3,000 samples spanning all four hosts, 15 years of time and several European countries. We have particularly large sample sets from East Anglia, Scotland, Sweden, and Germany. With a target of 50 genomes, we aim to focus on these four regions. Within each, we will select five samples associated unambiguously with coniferous trees and five from deciduous trees. We will also sequence samples from the three sister species: B. aereus, B. reticulatus and B. pinophilus. The resulting genomic sequences will be analysed to identify genomic regions showing greater similarity between samples from the same host than between samples from the same locality. Such genomic regions are indicative of a role in host adaption, predicting that the same regions will exhibit parallel patterns of divergence between B. edulis and its sister species.
