LUR1 Promotes Sortilin-induced Macrophage Lipid Accumulation and Aortic Atherosclerosis via Furin Maturation

Bioinformatics analysis was used to predict the interaction and correlation between LUR1 and Flynn protease, as well as their potential participation in the pathogenesis of atherosclerosis. Detect the binding of LUR1 to Flynn protein using immunoprecipitation assay. In THP-1 macrophages transfected with recombinant lentivirus, the expression of LUR1, Furin, pro Furin, Sortilin, and pro Sortilin was determined by RT qPCR and Western blotting. Quantify cellular lipid content using colorimetric method and observe intracellular lipid droplets through Oil Red O staining. Measure plasma lipid mass spectrometry using enzymatic oxidation method. Histological staining, immunofluorescence and immunohistochemistry were used to study aortic atherosclerosis and LDLR gene expression −/− mice fed a high-fat diet.
