Puccinia striiformis Westend f.sp. tritici Eriks. (Pst) causes stripe rust, an important disease of wheat worldwide. Recently, microRNA-like small RNAs (milRNAs) were discovered in several filamentous fungi, including many pathogens, but not yet in Pst. The latest reported that fungal small RNAs suppress plant immunity by hijacking host RNA interference pathways. Although RNA interference pathways such as quelling and meiotic silencing have been well described in Neurospora crassa, knowledge of smRNAs in fungi is still limited compared to other eukaryotes. In this study, to explore the potential milRNA in Pst that may regulate the processes of urediniospore germination and the role of pst sRNAs in pathogenicity, sRNA libraries of urediniospore germination were generated and sequenced using Solexa sequencing technology.
