We constructed a Bacterial Artificial Chromosome (BAC) library from the genomic DNA of Yersinia pestis antiqua strain 910706. The library was generated by cloning Hind III-digested fragments into the CopyControl pCC1BAC vector. It consists of 2,624 clones with an average insert size of 105 kb, providing approximately 8.3-fold coverage of the haploid genome. This library was utilized to identify and functionally characterize a 33 kb prophage-like DNA fragment (YPO2095-YPO2135), demonstrating that this region is not essential for the full virulence of Y. pestis. This BAC library serves as a valuable genomic resource for further investigation into gene function, genomic evolution, and genetic mapping in Y. pestis.
