HMGA2 project, CRISPR screen

sgRNA whole genome library sequencing in OCI-AML5-Cas9 EKO library cells overexpressing HMGA-YFP or control YFP vectors. The goal of this experiment is to identify synthetic lethal and synthetic rescue sgRNAs with regard to HMGA2 overexpression in AML. OCI-AML5-Cas9 EKO library cells were infected with HMGA-YFP or control YFP vectors for 3 days and then treated with DOX for 7 days to induce Cas9 expression. Cells were further expanded for 7 days before DNA extraction. ID sequences of the sgRNAs were amplified by PCR and sequenced.
