Miscanthus sinensis Andersson (Poaceae) is considered to be one of the most key feedstock for bioethanol production due to its high yield, low input, and low environmental impacts. In this paper, PacBio Iso-Seq sequencing technology and Illumina HiSeq technology were simultaneously applied on the same samples from Miscanthus sinensis, providing the largest full-length transcriptome dataset with 408,801 non-redundant transcripts. Through the whole genome sequence alignment, sorghum and maize were identified to be the phylogenetically closest species with Miscanthus, thus indicated a high utility of sugarcane and sorghum genomes as references for Miscanthus. Furthermore, multiple functional annotation reveals a wealth of candidate molecules such as TF transcripts and alternative splicing events that potentially underlie the diverse biological processes as well as the regulatory networks of M. sinensis.
