Cas9 fused to VP16 repeats have been used in several systems of gene activation. Here we aimed to evaluate the undesirable effects of this system whether in presence or absence of gRNAS targeting a specific gene, CD34. Overall design: HEK293T cells were transfected with dCas9-VP160 in the presence or absence of 6 gRNAs targeting the promoter of CD34, and two technical replicates of each treatment as well as a control (non-transfected cells) were assayed using dirRNAseq. This was repeated with a second batch of cells.
