The histone methyltransferase ASH1L, which deposits histone 3 lysine 4 trimethylation marks (H3K4me3), contributes to global genome nucleotide excision repair (GG-NER) of cyclobutane pyrimidine dimers (CPDs), DNA lesions caused by UV radiation, in part by interacting with XPC protein, the initiator of the GG-NER reaction . We investigated the mechanism by which it does so by suppressing its methyltransferase activity and observing the subsequent impact, upon UV exposure, on H3K4me3 deposition, XPC binding to chromatin, and CPD excision.
