High-temporal-resolution transcriptome analysis of the anti-IgM-stimulated mouse B cells

B cell receptor (BCR) is essential in activating B cells and mediating the immune responses. Analyzing the BCR-triggered gene expression would help delineate the regulatory function of B cells. The mouse B cells were treated with anti-IgM mAb to mimic the BCR stimulation, and the genome-wide transcriptional responses were analyzed by using high-throughput sequencing at high-temporal resolution (24 time points over 6 hours). Overall design: The mouse splenic B cells were isolated and purified by depleting CD43+ cells. The purified B cells were stimulated with 10 µg/ml of anti-IgM. The gene expression profiles under the anti-IgM stimulation were measured every 15 minutes in the time span from 0 time to 6 hours by RNA-seq analysis using Illumina HiSeq platform. Control was set as the non-stimulated (0 time) cells. Biological replicate samples were analyzed.
