TNBC- cisplatin resistance metabolomics study

A cisplatin-resistant TNBC cell line was developed and characterized. An untargeted metabolomics study was performed using LC-MS/MS technology. The study was done with biological and technical replicates. QC pools were run in between to check the instrument variation. Primary data analysis and metabolite identification were done using compound discoverer 3.2 and statistical analysis was performed using metaboanalysis 5.0. The target metabolite validation was also done using Parallel Reaction Monitoring (PRM) based analysis.
本研究构建并鉴定了一株顺铂耐药的三阴性乳腺癌(Triple-Negative Breast Cancer, TNBC)细胞系。采用液相色谱-串联质谱(Liquid Chromatography-Tandem Mass Spectrometry, LC-MS/MS)技术开展非靶向代谢组学研究,实验设置生物学重复与技术重复,并穿插运行质控混合样本以监测仪器偏差。 采用Compound Discoverer 3.2完成原始数据分析与代谢物鉴定,使用metaboanalysis 5.0开展统计学分析;同时采用基于平行反应监测(Parallel Reaction Monitoring, PRM)的分析方法完成靶向代谢物验证。
