Decoding the Escherichia coli epitranscriptome with 004kit

ONT has recently launched its latest sequencing kit (SQK-RNA004) and flow cell (FLO-MIN004RA), which offer significantly improved yields, more stable current, and enhanced sequencing quality. In addition, several new all-context modification models have been introduced since the launch of the rna004_130bps_sup@v5.0.0 model, including m6A, pseU, A-to-I, and m5C. To evaluate the new model and investigate bacterial mRNA modifications, we followed a well-established pipeline and independently performed in vitro-transcribed (IVT) RNA libraries as negative controls. This approach yielded high-quality data characterized by very stable expression levels, particularly for the samples ss&rd_004 and IVT_neg_004.
