Evaluation of tofacitinib in cutaneous sarcoidosis [scRNA-seq]

Sarcoidosis is a multisystem inflammatory disease and no molecularly targeted therapies exist. Here we evaluate the efficacy of tofacitinib (a JAK1/3 inhibitor) in an open-label trial of 10 patients with sarcoidosis and correlate clinical improvement with molecular parameters in blood and skin. Overall design: 4 mm punch biopsies of skin were obtained on separate days from 3 patients with GA and 3 healthy volunteers. Biopsies were immediately dissociated using dispase (10 mg/mL) followed by liberase. Live single cells were purified by FACS and single cell libraries were prepared using a 10X chromium controller according to the manufacturer's instructions and sequenced using an Illumina Hiseq 4000.
