PAX3 promotes melanoma progression by enhancing cell growth, migration, and survival while blocking differentiation. Its known targets are limited, and the roles of its two DNA-binding domains (PD and HD) are unclear. Using CUT&RUN in SK-MEL-5 cells, we identified PAX3-bound regions, categorized by motif and chromatin state. Most binding occurred through the PD at active sites, with minimal repressive activity. Pathway analysis linked targets to transcription, RNA processing, and neuronal function. These findings reveal new PAX3 targets and suggest that PD binding plays a key role in gene activation in melanoma.
