The goal of this project was to examine microbial diversity in high-altitude freshwater lakes in Yosemite National Park, CA, USA. Samples were collected from 15 lakes spread across an elevation gradient, with a subset of lakes sampled at several time intervals. DNA was extracted from filtered water samples, and archaeal/bacterial 16S rRNA genes were amplified and then sequenced (using Illumina).
