The heterogeneity of glioblastoma, an incurable brain tumour, facilitates parts of tumour cells evasion from tumour therapies, and leads to tumour relapse eventually. We created synthetic super-enhancers (SSEs) that focusing on the core SOX2 and SOX9-driven transcriptional regulatory network, thereby boosting the overall activity through transcription factor (TF) clustering while maintaining glioblastoma stem cells (GSCs)? selectivity via the natural enhancer grammar. Here, we revealed SSEs integrate the activities of SOX driven cell type identity, with transcriptional effectors of known GBM signalling pathways using single cell RNA sequencing analysis.
