The results of self-incompatible S haplotype identification showed that the cabbage "325" contained a Class II S haplotype. PCR amplification and sequencing analysis of SRK and SCR genes at the S locus showed that the cabbage "325" was inconsistent with the known Class II S haplotype. This indicates that the material contains a new S haplotype named BrS-325. Genetic analysis showed that the variation of a single S locus resulted in self-compatibility. In order to obtain the complete genomic information of Chinese cabbage "325" and analyze the reason of its self-compatibility, the whole genome of "325" was sequenced by nanopore sequencing technology. This genome can not only quickly obtain the S-locus sequence information of BRS-325, but also has important value in the comparative genome of other traits. To provide a good reference genome for basic research of Chinese cabbage.
