A benchmark of in-house single-molecule sequencing system (in-house HeliScope)

DNA barcodes are being leveraged for spatial transcriptomes, which are attracting attention as methodologies that provide information lacking in conventional transcriptomes. Although spatial resolution relies on methods used to decode DNA barcodes, achieving single-molecule decoding remains a challenge. Therefore, we developed a sequencing system to spatially decode DNA barcode molecules at single-molecule resolution without involving the amplification process, by repurposing a single-molecule sequencing system (HeliScope). Here we report a benchmark of in-house single-molecule sequencing system. For benchmarking, we analyzed a minute amount of cDNA library, comparable to that obtained from a single-cell, and compared them using an Illumina sequencer.
