Calcium dynamics drive metabolic regulation of epigenetic reprogramming at fertilization and alter EGA resulting in long term effects in offspring. A mouse model (C57B6) of dramatically increased calcium exposure at fertilization in vivo was developed by targeting two plasma membrane calcium pumps, isoforms 1 and 3 (PMCA1 and PMCA3) in mouse oocytes. dKO or Control females were mated to WT males. The liver, adipose tissue and muscle of offspring derived from dKO or control dams was assessed for differences in the methylation landscape of DNA.
