We investigated the cellular and transcriptional trajectories of adult neurogenesis in the sea anemone Nematostella vectensis through single-cell transcriptome analysis of Elav>KikGR+, FoxL2>KikGR+, SoxC>KikGR+ body wall with mesenteries (no oral region) adult samples. Overall design: Adult sea anemones (Nematostella vectensis) with a transgenic photoconvertible reporter (either Elav>KikGR, SoxC>KikGR, or FoxL2>KikGR) were photoconverted. At 2, 4 or 7 days post-photoconversion, animals were amputated below the pharynx and the body wall with mesenteries region was dissociated into single cells, KikGR+ cells were sorted using Fluorescence-Activated Flow Cytometry (FACS) into individual wells of 384-well plates. Single cell RNA-Seq libraries were generated with the MARS-Seq protocol (one scRNA-Seq library for each half-plate). Each sample here is one of 84 half-plate scRNA-Seq library.
